Adding PTMS to NMR chain

Is it possible to add ptms to my residues in the nmr chain?

e.g a phosphoserine/threonine instead of the standard amino acid.

Yes!

You will essentially need to create a Chain with non-canonical Amino Acids (the ones with the PTMs). There is information including a video (which uses Phosphoserine as an example!) on all of this at Creating Chains with Non-canonical Amino Acids

You will need a so-called ChemComp file for the atoms with PTMs. If your PTMs are fairly standard, then you should be able to find these ChemComp files on our GitHub page:

You will need to know what the PDB code for your PTM AA is. Probably the easiest way to find this is to Google something like “phosphotyrosine pdb chemical component” which would give you this as your top hit:

showing that the PDB code for phosphotyrsine is PTR which you can then pop in the search box on the GitHub page to get the ChemComp file.
